The thesis explores the crucial role of uncomplexed cationic polymer chains in enhancing gene transfection, challenging the established "proton sponge" model. Through a blend of synthetic chemistry, polymer physics, and molecular biology, it reveals that these free polymer chains, rather than those bound to DNA, significantly influence intracellular trafficking. The findings indicate that chains longer than 10 nm can obstruct fusion between endocytic vesicles, offering fresh perspectives for designing advanced polymeric gene-delivery vectors.
